My game is based off a book I wrote too. I hunted for files from other people's games and online resources before starting my project. That gave me a quick wealth of tilesets and charsets to pick from. Choose some tilesets that look most pleasing to you, and ones that look easy to work with. My next step was making maps and the database. Have fun by making your world come alive! Worry about the character appearance last, since you can always change it along the way. As for files to assist making your character sprites when the time comes, maybe someone else has suggestions.

My only tidbit of advice would be to learn a bit about creating custom charactersets if you can. You don't need much artistic talent when it comes to such small sprites; and even beginning somewhere as short as recolours will do.
One of my ongoing projects concerns a book I wrote, and my only major problem was resources, since you already have the plot to back you up. Try and find some interesting ways to introduce your characters visually. Especially considering it derives from written text already, all you have to do is visualise it for people.
My one major thing you should be careful about is trying to apply the actual gameplay aspects to it. A common problem in games based on stories is that the 'cut-scenes' are too long and there is little gameplay content. Think about which areas your story could have player interaction and proceed from there.

Just keep in mind books and games are different from each other. I've been working with SBester on writing a novel to his Eden Legacy series, which the book has wound up being completely different in a lot of parts do to more freedom and writing the absence of game play. In game, there's a more heavier focus on game play and dialogue, whereas in books on description and action.

It all depends on what you want to do. If it makes perfect sense to do so, it might work. But you must remember how close you want to stay to the original work. Establish a standard and stick to it.
Basically, it doesn't matter how close you get, just that you have consistency with your implementation.

Absolutely, based on just means most of the plot or story was taken from or originates around the book, there can still be some changes I.E the character. Just make sure the game and book share a similar story or plot. A few character changes won't really make the book stand apart from the game.

Even if you wanted your character to look exactly as described in the story, the major advantage of modern programming practices (which you don't need to know about) allow you to quickly swap in your desired character halfway or even after completing the game, and it would take 5 seconds to do so.
So you can start building the game and worry about very specific details later.
