Would it be possible to do that while on the main map? Stand around, maybe get a blink every few [random number here] seconds instead of the soulless stare?

Sure, assuming that you have the sprites for it.

You could have a parallel process event wait for a while, and then if no movement keys were being pressed, use Show Move Route to do some animation.

It'd require a chunk of fine-tuning, of course.

Absolutely. As I said, it'll probably take tweaking. You might need more than one event-- I'm not sure.
